We did a similar analysis recently on the interaction between ritonavir/darunavir, published in JAC by Stein Schalkwijk

It is hard to separate covariance in inter-individual variability from the drug interaction. You’ll have to make some assumptions or have data on both drugs without the interaction present.

Happy to share my thoughts if necessary.

Dear colleagues,

We are working on a model to describe a pharmacokinetic drug-drug interaction between two drugs.
I recall that in the past there has been a discussion on the forum about simultaneously modelling two different drugs for the sake of sharing covariates or covariate models between them.
Beyond those posts and a paper by van der Laan (AAC 2018) on a lopinavir-ritonavir DDI, I cannot find published (coding) examples of how to approach DDIs in a joint PK model.
Hopefully some of you are willing to point us into the right direction?
